The drug has a hypotensive effect. It dilates arterioles and thus reduces peripheral vascular resistance, disrupts the transmembrane transition of calcium ions into the cell through the LCM channels and reduces the flow of calcium into the smooth muscle cells of arterioles. Amlodipine reduces the tone of the smooth muscles of arterioles, which leads to vasodilation. In this case, there is a significant decrease in total peripheral vascular resistance and myocardial oxygen consumption, even in healthy people. The effect of amlodipine is mainly dose dependent. Amlodipine does not affect the activity of the sinus node, does not cause reflex tachycardia after coronary occlusion, and prevents the development of LV hypertrophy.
