The mechanism of action of chlorthalidone is that it blocks sodium reabsorption in the distal renal tubules. This causes increased excretion of sodium and water, as well as chloride, potassium, magnesium and calcium.

Chlorthalidone is usually prescribed at a dose of 25-50 mg once daily in the morning. The maximum daily dose is 100 mg. The drug is well absorbed from the gastrointestinal tract, reaching peak plasma concentrations after 2-4 hours. The half-life is 40-60 hours.
